Yes. A valid declaration requires at least one pure sequence. An impure sequence (with a joker) does not satisfy the requirement.

When should I drop?

Drop when your hand is weak after the initial draw and you cannot build a pure sequence within the first few picks. A first drop costs a fixed penalty that is usually smaller than the points you would carry.
